The main effects and Clomiphene pharmacodynamics:
Clomiphene is an antiestrogen of non-steroidal structure. Acts by specific binding with estrogen receptors of the ovaries and pituitary gland. In low concentrations of estrogen in the body, Clomiphene shows a moderate estrogenic effect, in high concentrations it has an antiestrogenic action. In small doses, Clomiphene increases the secretion of gonadotropins (prolactin, FSH and LH), stimulates ovulation, and in large doses - inhibits the secretion of gonadotropins. Does not express gestagenic and androgenic activity.
Proposed clinical use:
- anovulatory infertility (ovulation induction)
- dysfunctional metrorrhagia
- amenorrhea, secondary amenorrhea, post-contraceptive amenorrhea
- galactorrhea (in the background of pituitary tumor)
- polycystic ovary syndrome
- Chiari-Frommel syndrome
- androgenic insufficiency
- oligospermia
- testosterone level normalization
Absolute and relative contraindications: The drug must not be used in cases of:
- Hypersensitivity to the components of the preparations or a known allergic reaction to drugs of the same class.
- liver and / or renal failure
- metrorrhagia of unknown etiology
- ovarian cysts
- genital neoplasms
- tumor or hypofunction of pituitary gland
- endometriosis
- ovarian failure on the background of hyperprolactinemia
- pregnancy
The preparation should be used with caution after a consultation with your doctor in the following conditions:
- tendency to thrombosis
- bleeding
Possible adverse reactions: As any drug Clomiphene may express specific side effects. If a side effect is stated below it will not necessarily appear during the use of the preparation. However if any of the given side effects do manifest please consult your doctor immediately. Clomiphene adverse effects include the following:
- nervous system: dizziness, headache, drowsiness, slow mental and motor reactions, depression, irritability, insomnia
- digestive system: nausea, vomiting, gastralgia, flatulence, diarrhea
- allergic reactions: rare - rash, allergic dermatitis, vasomotor disturbances
- urogenital system: rare - polyuria, increased frequency of urination, pain in lower abdomen, dysmenorrhea, menorrhagia, increased size of the ovary (including cystic)
- other: weight gain, hot flush, rare - impaired vision, alopecia, breast tenderness
